Role of mitogen-activated protein kinases in the regulation of paraventricular nucleus to gastric ischemia-reperfusion injuries被引量:19

Background We investigated the role in electrical stimulations of paraventricular nucleus (PVN) on gastric mucosal cells and the activity of mitogen-activated protein kinases (MAPKs) family members induced by gastric ischemia-reperfusion (GI-R). And we elucidated the molecular mechanisms of the protection of PVN from GI-R injuries. Methods Sprague-Dawley rats were divided randomly into 4 groups: Group I, the sham-operated GI-R control group; Group II, the sham-operated electrical stimulations to PVN + sham-operated GI-R control group; Group III, the GI-R group; and Group IV, the electrical stimulations to PVN + GI-R group. In all of the experiments, the PVN was stimulated prior to the induction of GI-R. The GI-R model was established by clamping the celiac artery for 30 minutes to induce ischemia and then was released to allow reperfusion for 30 minutes, 1 hour, 3 hours and 6 hours, respectively. The gastric mucosal cellular apoptosis, proliferation, and the expression and activity of MAPKs protein were observed by immunohistochemistry and Western blotting, respectively. Results Compared with the GI-R group, the application of electrical stimulations in the PVN significantly depressed gastric mucosal cellular apoptosis and enhanced gastric mucosal cellular proliferation following the 30-minute, 1-hour and 3-hour intervals of reperfusion; it also promoted the activation of p-ERK during the early phase of reperfusion but inhibited the activation of p-JNK1/2 and p-p38 following the 30-minute, 1-hour and 3-hour intervals of reperfusion. Conclusions The protection of PVN against GI-R injuries may attribute to the inhibition of apoptosis and the promotion of the proliferation of gastric mucosal cells during GI-R. This protective effect is mediated by activating the ERK pathway and depressing the JNK, the JNK. p38 MAPK oathwavs of the oastric mucosal cells.

Hypothalamic paraventricular nucleus(PVN) is an important nucleus of regulating viscera-functions,which contains many transmitters and projects extensively.PVN and its peptidergic neurons take part in regulating various activities of the stomach,e.g.its secretion,electrical activation and gastric motor.Fos-immunoreactive neurons in the PVN were largely expressed after gastric noxious stimuli.The activation of neurons in PVN takes part in regulating various gastric mucosal injuries via the mediation of vagus and sympathetic nerve or neuroendocrine pathways to change the aggressive factors and defensive factors in gastric mucosa.

吡咯烷二硫氨基甲酸酯对大鼠胃缺血/再灌注损伤的影响

目的研究核因子κB(NF-κB)抑制剂吡咯烷二硫氨基甲酸酯(PDTC)对大鼠胃缺血/再灌注(gastricischemia/reperfusion,GI/R)损伤的影响。方法采用夹闭大鼠腹腔动脉30 min,去除动脉夹再灌注1 h的GI/R损伤模型,于静脉注射PDTC后,肉眼计数胃黏膜损伤指数,并运用免疫组化及TdT介导dUTP缺口末端标记(TUNEL)实验技术,观察PDTC对大鼠GI/R后胃黏膜细胞坏死、增殖和凋亡的影响。结果夹闭大鼠腹腔动脉30 min再灌注1 h后可造成胃黏膜明显损伤,静脉注射PDTC 200 mg/kg可明显减轻GI/R损伤,并使胃黏膜细胞增殖增加、凋亡减少。结论静脉注射PDTC对GI/R损伤具有保护作用,这种保护作用是通过促进胃黏膜细胞增殖、抑制胃黏膜细胞凋亡而实现的。

下丘脑室旁核内注射谷氨酸对内脏高敏感性大鼠内脏痛的影响及机制被引量:3

目的研究下丘脑室旁核(hypothalamic paraventricular nucleus,PVN)内注射谷氨酸(glutamate,Glu)对慢性内脏高敏感性(chronic visceral hypersensitivity,CVH)大鼠慢性内脏痛的影响及其可能的机制。方法新生SD大鼠,在出生后第8、10、12天分别给予结直肠扩张法制备CVH大鼠模型。取30只制备成功的CVH模型大鼠,随机分为CVH模型组(CVH组)、CVH+PVN内注射生理盐水组(NS组)、CVH+PVN内注射Glu组3、6、12 μg组(G3组、G6组、G12组),每组6只,另取体质量相匹配的6只SD大鼠作为假手术组(Sham组)。采用痛阈、腹壁撤退反射(abdominal withdrawal reflex,AWR)评分和腹外斜肌肌电图(abdominal external oblique muscle electromyography,EMG)评价大鼠的痛行为。免疫组化染色法检测大鼠脑内精氨酸加压素(arginine vasopressin,AVP)表达水平和结肠组织细胞增殖情况,Tunel法检测结肠组织细胞凋亡情况。结果与NS组比较,G3、G6、G12组大鼠的痛阈均升高,AWR评分和EMG幅度均降低,差异有统计学意义[痛阈:t=7.65,16.31,24.78,均P<0.05;AWR评分:t=-2.98,-4.77,-7.29,均P<0.05;EMG幅度:t=-3.06,-5.75,-8.92,均P<0.05]。与Sham组[(42.63±5.20)%]比较,CVH组和NS组大鼠PVN内AVP表达下降[(18.67±2.94)%,(17.53±2.47)%;t=6.95,7.56,均P<0.05],大鼠PVN内注射不同剂量Glu后,AVP表达升高,其中G12组为[(37.03±7.63)%],与NS组比较,差异有统计学意义(t=-4.21,P<0.05)。与Sham组[(65.48±1.68)%]比较,CVH组和NS组大鼠结肠组织黏膜细胞增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)表达降低[(18.39±1.67)%,(17.69±1.68)%;t=34.35,34.80,均P<0.05],PVN内注射不同剂量Glu后,PCNA表达升高,其中G12组为[(59.91±5.63)%],与NS组比较,差异有统计学意义(t=-12.44,P<0.05)。与Sham组[(23.38±11.40)%]比较,CVH组和NS组大鼠结肠组织黏膜凋亡细胞表达升高[(83.79±3.57)%,(80.91±2.47)%;t=-8.77,-8.54,均P<0.05],PVN内注射不同剂量Glu后,凋亡细胞表达降低,其中G12组为[(18.15±6.49)%],与NS组比

茉莉酸甲酯对胃癌SGC7901细胞生长抑制机制探讨被引量:2

目的:探讨茉莉酸甲酯(MJ)对胃癌SGC7901细胞生长的抑制作用及其机制。方法:四甲基偶氮唑(MTT)法检测细胞生长活性;Hoechst 33258荧光染色法观察凋亡细胞核的形态学变化;AnnexinⅤ-FITC/PI双染流式细胞术检测细胞的凋亡率;蛋白质印迹法检测凋亡蛋白Caspase-3和Caspase-9的表达。结果:MTT法显示,MJ对胃癌SGC7901细胞生长有显著抑制作用,且呈剂量-时间依赖关系,0.5、1.0和2.0mmol/L MJ作用6h后,抑制率分别为(7.10±0.84)%、(18.22±1.82)%和(26.89±1.56)%,溶剂对照组为(0.43±0.26)%,顺铂(DDP)组为(10.44±1.17)%,总体比较差异有统计学意义(F=232,P=0.000),各组间比较差异均有统计学意义,P值均<0.01;采用MJ上述3种浓度作用12h后,抑制率分别为(12.44±1.20)%、(32.70±1.29)%和(52.94±2.29)%,溶剂对照组为(0.31±0.41)%,DDP组为(22.60±1.97)%,总体比较差异有统计学意义(F=1 013,P=0.000),各组间比较差异均有统计学意义,P值均<0.01;MJ三种浓度作用24h后,抑制率分别为(17.37±1.95)%、(41.79±4.40)%和(71.93±4.65)%,溶剂对照组为(0.34±0.21)%,DDP组为(27.72±2.45)%,总体比较差异有统计学意义(F=444,P=0.000),各组间比较差异均有统计学意义,P值均<0.01;其中以浓度为2.0mmol/L MJ作用24h后,对SGC7901细胞的抑制率效果最佳。荧光染色法观察MJ能够诱导人胃癌SGC7901细胞株的凋亡。流式细胞检测显示,MJ可促进胃癌SGC7901细胞的凋亡,凋亡率为(68.14±7.91)%;与溶剂对照组(6.98±1.45)%相比,差异有统计学意义,P=0.013。蛋白质印迹法检测显示,MJ可上调凋亡蛋白Caspase-3和Caspase-9的表达;事先采用辣椒素受体拮抗剂(Cap)预处理,MJ的抗胃癌SGC7901细胞的效应可被消除,与MJ单独给药组相比,MJ对癌细胞的生长抑制率下降为(38.79±2.77)%,P=0.05;凋亡细胞明显减少;肿瘤细胞凋亡率降为(39.81±4.57)%,P=0.045;凋亡蛋白Caspase-3和Caspase-9表达也降低。结论:MJ有显著抑制胃癌SGC7901细胞增殖的作用;辣椒素�
