The association of LOXL1 polymorphisms with exfoliation syndrome/glaucoma: Meta-analysis被引量:1

AIM: To investigate the association of lysyl oxidaselike 1(LOXL1) single nucleotide polymorphisms(SNPs)with exfoliation syndrome(XFS)/exfoliation glaucoma(XFG).METHODS: Published manuscripts from Pub Med and EMBASE were identified until May 2014. Summary odds ratios(ORs) and 95% confidence intervals(CIs) for LOXL1(rs1048661, rs2165241 and rs3825942) polymorphisms and the risk of XFS/XFG were estimated using random-or fixed- effect model.· RESULTS: The three LOXL1 polymorphisms(rs1048661, rs3825942, and rs2165241) were associated with an increased risk for XFS/XFG among Caucasians,with OR 2.19(1.96-2.45), 8.8(6.05-12.79) and 3.41(3.11-3.73), respectively. On the contrast, the rs1048661 and rs2165241, but not rs3825942 polymorphism, have a potential protective effect on XFS/XFG in Asians, with OR0.06(0.02-0.18), 0.15(0.09-0.25), respectively.CONCLUSION: There is strong evidence that LOXL1 polymorphisms are associated with XFS/XFG risk. The strength of risk might be ethnicity-dependent.

叶黄素抑制叔丁基过氧化氢诱导的视网膜神经节细胞凋亡被引量:4

目的:观察叶黄素(lutein)对叔丁基过氧化氢(t-BHP)处理的视网膜神经节细胞(RGC-5细胞系)的保护效应并探讨其作用机制。方法:用免疫荧光染色检测视网膜神经节特异性蛋白Brn-3和神经微管结合蛋白MAP-2的表达来鉴定RGC-5细胞;将RGC-5细胞随机分为对照组、t-BHP处理组、t-BHP和lutein共同处理组、lutein处理组,培养24 h,MTT实验检测细胞活力;Annexin V-FITC/PI双染流式细胞术检测细胞凋亡;免疫细胞化学技术检测caspase-3蛋白的活化情况;Western blot检测Bcl-2/Bax、cleaved caspase-3、JNK和c-Jun蛋白的变化。结果:MTT实验和流式细胞检测结果显示,lutein能提高t-BHP处理的RGC-5细胞的活力,并降低t-BHP诱导的RGC-5细胞凋亡;免疫荧光结果显示lutein能抑制t-BHP诱导的caspase-3的活化;与对照组比较,t-BHP处理后RGC-5细胞抗凋亡蛋白Bcl-2表达下调(P<0.05),Bax/Bcl-2比率升高,cleaved caspase-3表达上调(P<0.05),JNK和c-Jun蛋白的磷酸化水平增加(P<0.05),t-BHP的上述作用可被lutein部分逆转。结论:Lutein能够降低t-BHP诱导的RGC-5细胞凋亡,其机制与其上调Bcl-2的表达、抑制caspase-3的活化并降低JNK和c-Jun蛋白的磷酸化有关。

重组蛋白PTD-HSP27的制备及其穿细胞膜和角膜组织的功能研究被引量:5

目的:制备PTD-HSP27蛋白并研究其能否穿过人晶状体上皮细胞SRA01/04细胞膜及新西兰大白兔的角膜组织进入眼前房。方法:利用重叠延伸PCR法获得重组目的基因片段PTD-HSPB1-6His,构建重组质粒p KYB-PTD-HSP27-6His,原核诱导PTD-HSP27蛋白表达及纯化重组蛋白后对其进行Western blotting鉴定;用异硫氰酸荧光素(FITC)标记重组蛋白PTD-HSP27并检测其穿透SRA01/04细胞膜的能力及兔眼角膜组织的能力。结果:成功构建并纯化制备目的蛋白PTD-HSP27,在PTD-HSP27孵育后的SRA01/04细胞及结膜囊滴注PTDHSP27兔眼前房内均可检测出重组蛋白PTD-HSP27。结论:通过重叠延伸PCR法及镍柱层析纯化法可以获得较纯的重组蛋白PTD-HSP27;重组蛋白PTD-HSP27能够穿透SRA01/04细胞膜及穿越兔眼角膜进入房水。

远志皂苷元对氧化应激损伤的视网膜神经节细胞的保护作用被引量:7

目的:观察远志皂苷元(senegenin,Sen)对氧化应激损伤的视网膜神经节细胞(retinal ganglioncells,RGCs)的影响并初步探讨其作用机制。方法:采用上丘荧光金逆行标记RGCs后,体外原代RGCs混合细胞培养,随机分为control组、H2O2组、Sen+H2O2和Sen组。检测带荧光金荧光细胞的活力。同上述分组处理视网膜,用Hoechst 33258染色后观察视网膜细胞核形态的变化,Western blotting检测视网膜细胞cleaved caspase-3、细胞色素C及Bcl-2蛋白的表达。结果:与control组比较,Sen浓度在10、20和40μmol/L时,RGCs活力无明显变化(P>0.05),但Sen浓度达到80和160μmol/L时,RGCs活力明显下降,差异显著(P<0.01)。25、50、100和200μmol/L H2O2明显降低RGCs活力(P<0.05)。Sen浓度在10、20和40μmol/L时,对50μmol/L H2O2损伤的RGCs有较好的保护作用(P<0.05),其中40μmol/L Sen保护作用最为明显。Hoechst 33258染色表明Sen可以减少H2O2引起的视网膜细胞凋亡。Western blotting结果表明Sen促进Bcl-2蛋白的表达,降低线粒体细胞色素C的释放,下调cleaved caspase-3的表达。结论:Sen保护RGCs对抗氧化应激引起的损伤,其机制可能与其增强Bcl-2蛋白表达和减少氧化应激引起的细胞凋亡有关。

Effects of Senegenin against Hypoxia/Reoxygenation-Induced Injury in PC12 Cells被引量:3

Objective:To investigate the effect and the potential mechanism of Senegenin(Sen) against injury induced by hypoxia/reoxygenation(H/R) in highly differentiated PC12 cells.Methods:The cultured PC12 cells were treated with H/R in the presence or absence of Sen(60 μmol/L).Four groups were included in the experiment:control group,H/R group,H/R+Sen group and Sen group.Cell viability of each group and the level of lactate dehydrogenase(LDH) in culture medium were detected for the pharmacological effect of Sen.Hoechst 33258 staining and annexin V/propidium iodide double staining were used to analyze the apoptosis rate.Moreover,mitochondrial membrane potential(△Ψm),reactive oxygen species(ROS) and intracellular free calcium([Ca^(2+)]i) were measured by fluorescent staining and flow cytometry.Cleaved caspase-3and activity of NADPH oxidase(NOX) were determined by colorimetric protease assay and enzyme linked immunosorbent assay,respectively.Results:Sen significantly elevated cell viability(P<0.05),decreased the leakage of LDH(P<0.05) and apoptosis rate(P<0.05) in H/R-injured PC12 cells.Sen maintained the value of△Ψm(P<0.05) and suppressed the activity of caspase-3(P<0.05).Moreover,Sen reduced ROS accumulation(P<0.05) and[Ca^(2+)]i increment(P<0.05) by inhibiting the activity of NOX(P<0.05).Conclusion:Sen may exert cytoprotection against H/R injury by decreasing the levels of intracellular ROS and[Ca^(2+)]_i,thereby suppressing the mitochondrial pathway of cellular apoptosis.

人参皂苷Rg1抑制叔丁基过氧化氢诱导的原代大鼠皮层神经元损伤被引量:4

目的:观察人参皂苷Rg1对叔丁基过氧化氢(t-BHP)诱导的原代大鼠皮层神经元损伤的改善作用,并探讨其可能机制。方法:将神经元随机分为正常对照组、10μmol/L t-BHP组及10μmol/L t-BHP+10μmol/L人参皂苷Rg1组,培养24 h。采用MTT检测不同浓度t-BHP处理的神经元活性,神经元三维重建研究神经元平均总纤维长度及总突起数量,免疫荧光检测caspase-3表达水平及免疫印迹方法检测Bcl-2、caspase-3以及磷酸化糖原合成酶激酶3β(pGSK-3β)蛋白表达水平。结果:10μmol/L人参皂苷Rg1能够对抗10μmol/L t-BHP引起的原代大鼠皮层神经元活性水平的降低,并且上调Bcl-2及pGSK-3β蛋白表达量,降低caspase-3活化为cleaved caspase-3的水平(P<0.05)。结论:人参皂苷Rg1可能通过提高GSK-3β自身磷酸化从而增强神经元的抗t-BHP损伤能力。

虹膜拉钩在白内障伴晶状体半脱位超声乳化手术中的应用被引量:4

目的:观察虹膜拉钩在白内障伴晶状体半脱位超声乳化手术中的应用.方法:17例白内障伴晶状体半脱位患者行白内障超声乳化手术,术中使用虹膜拉钩固定晶状体前囊膜,植入3片式人工晶体.结果:患者术后1d、1周、1个月及3个月术眼裸眼视力分别为0.47±0.21、0.51±0.23、0.56±0.24、0.52±0.27,最佳矫正视力分别为0.53±0.23、0.56±0.21、0.57±0.31、0.57±0.29,术后3个月13例人工晶体位置居中.结论:虹膜拉钩在白内障伴晶状体半脱位超声乳化手术中的应用明显降低手术风险,值得推广应用.

中药及有效成分对视神经损伤后视神经节细胞的保护作用
2011年
视神经损伤可由多种因素引起.如外伤、青光眼、炎症、缺血及肿瘤等。损伤导致视网膜神经节细胞(retinal ganglion cells,RGCs)继发性变性,是不可逆性视觉功能障碍的主要原因。损伤后短期内控制RGCs坏死和抑制RGCs继发性凋亡及促进受损节细胞神经轴突的再生成为视觉恢复的关键因素。某些中药在抗氧化、抑制自由基产生、扩张血管、降低外周血管阻力、改善微循环等方面有比较满意的效果.这些作用对视神经损伤后的保护十分关键。本文将对中药及有效成分在这方面的作用进行综述。

Comparison of trabeculectomy and trabeculectomy with amniotic membrane transplantation in the same patient with bilateral glaucoma被引量:4

AIM:To observe effects of trabeculectomy with amniotic membrane transplantation(AMT) in controlling postoperative intraocular pressure(IOP) in patients with medically uncontrolled glaucoma.·METHODS:This study included adult patients with requiring bilateral glaucoma surgery.Each patient underwent trabeculectomy(Non-AMT group) in one eye and with AMT(AMT group) in the other eye according to randomized principle.Success was defined as intraocular pressure(IOP) <21mmHg without any anti-glaucoma medications at 24 months follow-up.The two groups were compared in terms of IOP,complications and success rate.·RESULTS:Thirty-four eyes of 17 patients were investigated in this study.There was no statistically signifcant difference in pre-operative IOP between the two groups.The mean IOP was lower in AMT group compared with Non-AMT group on follow up months 12,18,and 24.Postoperative complications were more frequent in Non-AMT group(35.3%,6/17) compared with AMT group(5.9%,1/17).The success rate of surgery was88.2%(15/17) in Non-AMT group and 100%(17/17) in AMT group.·CONCLUSION:Trabeculectomy with AMT is an effective procedure to reduce IOP and complications,thereby improving surgical success rates.

AIM: To investigate the protective effect and its mechanism of lycium barbarum polysaccharides(LBP)against oxidative stress-induced apoptosis in human retinal pigment epithelial cells.METHODS: ARPE-19 cells, a human retinal pigment epithelial cell lines, were exposed to different concentrations of H2O2 for 24h, then cell viability was measured by Cell Counting Kit-8(CCK-8) assay to get the properly concentration of H2O2 which can induce half apoptosis of APRE-19. With different concentrations of LBP pretreatment, the ARPE-19 cells were then exposed to appropriate concentration of H2O2, cell apoptosis was detected by flow cytometric analysis. Expression levels of Bcl-2 and Bax were measured by real time quantitative polymerase chain reaction(RT-PCR) technique.RSULTS: LBP significantly reduced the H2O2-induced ARPE-19 cells' apoptosis. LBP inhibited the H2O2-induced down-regulation of Bcl-2 and up-regulation of Bax.CONCLUSION: LBP could protect ARPE-19 cells from H2O2-induced apoptosis. The Bcl-2 family had relationship with the protective effects of LBP.
