Silica hollow spheres(SHSs) have attracted great attention because of their low toxicity,low density,large surface area,high chemical and thermal stability,and surface permeability.They can be widely applied in storage[1],catalysis[2],drug delivery[3,4],low-dielectric-constant materials[5],low-refractive materials[6-8],and so on.Up to now,there have been various methods to produce SHSs.Inorganic[9] or organic particles[10],such as polystyrene or calcium carbonate,were used as hard templates to create hollow cavities.However,the multistep synthetic process and the lack of structural robustness of the shells upon template removal process weaken their application.Soft templates,including oil-in-water emulsions[1],[2],vesicles[13],micelle[14,15] and gas bubbles[16],are applied widely.

纳米粒子表面修饰对其与细胞膜相互作用的影响被引量:1

纳米粒子在生物医学上的应用越来越广泛,其进入细胞的机制与规律是设计与开发的基础.已有研究发现,表面修饰不同亲疏水性基团的金纳米粒子,在内吞机制被抑制时,进入细胞的能力明显不同.更特别的是,粒子表面亲水性基团与疏水性基团呈间隔条纹规则排列的纳米粒子,与其他修饰成分相同仅排列不同的纳米粒子进入细胞的规律区别显著.这一特殊现象无法用已有的纳米粒子进入细胞的机制解释.本文针对该研究结果,将纳米粒子与细胞的体系简化,定量分析了3种不同纳米粒子进入细胞前后的不同状态,计算获得了表面修饰不同亲疏水性基团的纳米粒子与细胞膜之间相互作用的Flory-Huggins自由能.结果发现,修饰规则间隔排列亲疏水基团的纳米粒子,其作用自由能在与细胞接触前后变化最大.研究结果不仅解释了实验发现,同时预示了纳米粒子进入细胞的新机制.

Study of alternate cooling and heating treatment induced tumor microvasculature injury被引量:1

Tumor microvasculature is important to tumor growth, metastasis and thus tumor treatment outcome. The alternate cooling and heating treatment has been confirmed to have advantages over single treatment of cooling or heating. The degree of tumor microvasculature damage induced by the alternate cooling and heating treatment and the mechanisms underlying are studied in this paper. The response of the tumor microvasculature to different treatments including alternate cooling and heating is observed in vivo through confocal microscopy using the nude mice dorsal skin fold tumor chamber model. Results show that alternate cooling and heating has induced much more severe damage to the microvessel structure throughout the entire tumor. Numerical simulations of the mechanical stresses on the tumor vessel wall has found that during the alternate treatment, the vessel wall suffer a rapid chang in thermal stresses in the opposite directions successively, which might caused damage to the peripheral microvasculature and micro-cracks in the central vessels. Reperfusion of blood flow after freezing also led to relatively large stresses on the vessel wall, especially when blood flow re-perfuses quickly during the subsequent heating. The quick increase of stresses on the blood vessel might be one of the key issues causing the blood vessel rupture referring to the experimental observation. The preliminary study has partly revealed the mechanism underlying serious tumor microvasculature damage caused by the alternate cooling and heating treatment.

冷热交替治疗对肿瘤微循环的损伤及机理研究被引量:1

肿瘤微循环对肿瘤生长和转移至关重要.冷热交替治疗已被证实优于以往的单一冷疗或热疗,其对于肿瘤微循环的损伤程度及机理是肿瘤成功治疗的关键,但目前该方向的研究甚少.利用裸鼠脊背皮翼肿瘤视窗模型结合激光共聚焦显微镜技术综合比较了单冷、单热及冷热交替对肿瘤微循环的损伤程度,并建立了分析肿瘤血管在热物理作用下受力的理论模型,通过数值计算获得了冷热交替治疗过程中肿瘤微循环血管壁的受力大小.研究结果发现,冷热交替造成了肿瘤微循环尤其是肿瘤中央成熟血管严重的结构性损伤.冷热交替过程中,血管壁先后受到迅速变化的方向相反的热应力,有可能在血管壁形成微小裂纹.随后加热造成的血流快速再灌注对血管壁的作用力存在应力集中现象,推测血液流动对肿瘤血管壁的快速冲击力很可能是造成血管壁破裂的关键因素,初步揭示了冷热交替治疗过程中肿瘤血管发生严重损伤的机理.

通过表面活性剂、共结构导向剂(CSDAs)和硅源的自组装合成了具有分散性的不同粒径氨基酸双功能化介孔二氧化硅纳米颗粒.利用表面活性剂头部与带相反电荷的CSDAs之间的静电相互作用使氨基和羧基基团均匀排列在介孔孔道表面.通过调节助溶剂或分散剂的加入量来控制颗粒粒径,调节合成溶液pH,改变纳米颗粒表面羧基基团和氨基基团的电荷量来控制颗粒的分散性.
