基于Biolog解析添加酶液对堆肥化过程中微生物群落代谢的影响被引量:8

以Biolog方法为主要监测手段,结合聚类分析以及PCA分析方法,考察了添加酶液对农业废物堆肥中木质纤维素降解及微生物群落代谢能力的影响.结果表明,添加酶液使得堆料中有机质的降解率提高了4.90%;微生物群落代谢聚类结果表明,添加酶液可以改善堆肥微生物对中间代谢产物类碳源的代谢能力.通过PCA发现,添加酶液提高了堆体中微生物对双亲化合物、聚合物、氨基酸和氨基化合物等碳源的代谢能力,由此可导致有机质被更高效降解;此外,堆肥各时期群落代谢聚类结果表明,酶液对堆肥进程的影响主要表现在一次发酵的第6 d和二次发酵的第30 d,从而有效地加速了堆肥进程.

农业有机废物与城市生活垃圾堆肥高温期微生物种群结构比较被引量:12

通过PCR、克隆文库方法分析了农业有机废物和城市垃圾堆肥高温期间细菌和真菌种群的多样性。提取堆肥高温期的DNA,PCR扩增,构建各高温期的16S rDNA和18S rDNA克隆文库,结果表明:农业有机废物和城市生活垃圾16S rDNA克隆文库中分别共有18个、21个OTUs,分别属于细菌域的14个、15个不同属,其18S rDNA克隆文库中分别共有8个、9个OTUs,分别属于细菌域的8个、9个不同属,推断农业有机废物堆体的优势菌为Bacillus megaterium、Rhizobium sp.、Phanerochaete chrysosporium、Penicillium sp.同属或同种的菌株;城市生活垃圾堆体的优势菌为Bacillus megaterium、Azospirillum sp.、Phanerochaete chrysosporium同种或同属的菌株。

简青霉(Penicillium simplicissimum)对木质纤维素的降解及相关酶活性特征被引量:4

简青霉[Penicillium simplicissimum(Oudem.)Thom BGA]能分泌木质纤维素降解酶,其中半纤维素酶、纤维素酶、木质素过氧化物酶、锰过氧化酶和漆酶的最大酶活分别为146.82 Iu·g-1、2.78 U·g-1、47.97 U·g-1、34.56 U·g-1和17.94 U·g-1.实验结果和SPSS统计分析表明,简青霉产木质纤维素酶的能力与木质纤维素的结构有很大的关系,其对木质纤维素的降解可能是几种木质纤维素酶之间协同作用的结果.在30 d的固态发酵中,半纤维素含量与发酵天数呈显著负相关(r=-0.946,P<0.01),纤维素与木质素的降解趋势呈显著负相关(r=-0.818,P<0.05).木质素过氧化物酶和锰过氧化物酶在降解木质素的同时对半纤维素和纤维素进行协同降解,是非选择性的木质素降解酶.木质素过氧化物酶、锰过氧化物酶与纤维素酶之间呈显著相关(相关性依次为r=0.922,P<0.01;r=0.807,P<0.05).研究还发现生物吸附在简青霉对液态碱木质素的去除中起到了非常重要的作用.

木质素降解酶在不同堆肥基质中的吸附传输特性研究被引量:2

为了深入探究木质素降解酶在不同堆肥基质中的吸附传输特性,通过批量实验对比了土壤、菜叶、稻草和米糠对木质素降解酶的吸附性能,并进行了动力学分析和等温吸附模型拟合,同时通过柱淋洗实验考察了木质素降解酶在4种堆肥基质中的迁移传输特性.结果表明,堆肥基质对木质素降解酶的吸附与基质种类有关,土壤、菜叶、稻草和米糠对木素过氧化物酶(LiP)和锰过氧化物酶(MnP)的吸附量分别为1.22、1.27、1.13、1.22U·g-1和5.09、4.88、4.43、3.95U·g-1.比较LiP和MnP吸附的动力学模型,准二级动力学方程为表征木质素降解酶吸附的最佳模型,其R2值为0.9732~0.9997,Elovich方程拟合较差,准一级动力学方程拟合最差;Langmuir模型对等温吸附数据进行拟合效果最好,而实验数据不适合用Freundlich方程表征.土壤、菜叶、稻草和米糠对LiP和MnP饱和吸附容量分别为1.23、1.30、1.17、1.14U·g-1和5.70、5.19、4.73、4.14U·g-1.LiP和MnP在稻草和米糠基质中传输效果较好,可传输到最深层10mL处,而在土壤和菜叶基质中则被滞留在浅层.

Spectroscopic studies of dye-surfactant interactions with the co-existence of heavy metal ions for foam fractionation被引量:2

The interaction between a cationic dye Methylene Blue(MB) and an anionic surfactant sodium dodecyl sulfate(SDS) with the presence of Cd2+ was investigated spectrophotometrically in a certain concentration range.The spectrophotometric measurements of dye-metal ion-surfactant system were carried out.The results indicated that the SDS concentration had a significant influence on the dye spectrum,while the addition of Cd2+ hardly caused change of the maximum value of absorbance.According to this observation,we concluded that electrostatic and hydrophobic interaction between dye and surfactant occurred up to a certain level,and the homo-ions Cd2+ almost exerted no effect on the dye-surfactant complexation,establishing a theoretical foundation for simultaneous removal of organic dye and heavy metal using foam fractionation.Meanwhile,the effects of their interaction on foam performance were investigated.The results showed that the addition of Cd2+ favored the tendency to ameliorate foam properties just contrary to MB.The feasibility of foam separation for dye and heavy metal removal from simulated wastewater was also confirmed using a continuous foam fractionator.In the simultaneous removal process,with the initial SDS concentration ranging from 0.5 to 5.0mmol/L,the maximum removal efficiencies of MB and Cd2+ were obtained as 99.69% and 99.61%,respectively.The enrichment ratios were reduced from24.34 to 7.65 for MB and from22.01 to 3.35 for Cd2+.

胞外酶对堆肥中微生物群落演替的影响被引量:5

研究了添加黄孢原毛平革菌胞外酶对农业废物堆肥中木质纤维素降解及微生物群落演替的影响.结果表明,堆肥初期,胞外酶对微生物生长具有抑制作用,且随堆肥时间的延长逐渐减弱;堆肥后期,胞外酶可提高微生物群落的多样性和均匀性,并促进木质素和半纤维素的大量降解.醌指纹数据显示,胞外酶并未引起新的优势醌的出现,进入高温期后,以MK-7为主要醌类的微生物作为优势菌群,且这种优势一直存在;堆肥后期胞外酶对甲基萘醌影响较大,可促进长链甲基萘醌的出现.主成分分析表明,胞外酶对醌种类演替的影响主要作用在3d和40d,这与非相似性指数结果一致,并使真菌在整个体系中影响力加大.

Transformation behavior of lead fractions during composting of lead-contaminated waste

The transformation behaviors of Pb fractions during composting of Pb-polluted waste without inoculants and with the inoculants of Phanerochaete chrysosporium were studied.Results show that the active Pb ions with high toxicity and transferability are transformed into the inactive Pb with low toxicity and transferability,confirming that Pb ions can be efficiently immobilized during composting without or with the inoculants.The soluble-exchangeable Pb in composting without inoculants reaches 49.0 mg/kg at day 60,while that with the inoculants is reduced to 0 mg/kg dry mass compost.The higher contents of organic-bound Pb (59.0 mg/kg)and residual Pb(69.2 mg/kg)with low toxicity are found after 60-day composting with inoculants,compared with those without inoculants.The above data indicate the better immobilization effect of Pb and the greater alleviation of Pb hazards in composting with the inoculants of Phanerochaete chrysosporium than without inoculants,which may be due to the more microbial biomass and the higher pH value in composting of Pb-polluted waste with inoculants.

愈创木酚与复合碳源共降解对黄孢原毛平革菌产酶的影响被引量:1

通过正交实验法,研究愈创木酚与复合碳源共降解对黄孢原毛平革菌产酶的影响,同时探讨不同分子结构对酶催化机制的影响。实验结果表明:同时添加愈创木酚及不同结构的碳源对该菌产酶有显著影响,高浓度的愈创木酚对产酶有明显的促进作用;在培养基中添加愈创木酚2 mmol/L、葡萄糖2.5 g/L和糊精5 g/L,可以显著提高黄孢原毛平革菌的综合产酶能力;黄孢原毛平革菌可以分泌纤维素酶和木聚糖酶,而且二者之间有较好的线性正相关关系。

Fluorescence spectroscopy characteristics of humic acid by inoculating white-rot fungus during different phases of agricultural waste composting被引量:2

The white-rot fungus, Phanerochaete chrysosporium (P. chrysosporium), was inoculated during different phases of agricultural waste composting, and its effect on the fluorescence spectroscopy characteristics of humic acid (HA) was studied. The results show that the emission spectra have a sharp peak at 400 nm and a broad shoulder with the maximum centered at 460 nm. The excitation spectra have two peaks and exhibit red shift (shift to longer wavelengths) at 470 nm. The synchronous scan spectra present a number of peaks and shoulders, and the peaks at shorter wavelengths disappear gradually and form a shoulder. At the final stage of composting, the fluorescence spectra have similar shapes, but the fluorescence intensities decrease. P. chrysosporium increases the degree of aromatization and polymerization of HA when it is inoculated during the second fermentation phase, while it does not produce an obvious change on the humification degree of HA when it is inoculated during the first fermentation phase. Compared with the fluorescence spectroscopy characteristics of HA from soil, the structure of HA from compost is simpler and the activity is higher.
