水稻RBX1基因cDNA的分离及其过量表达载体对水稻的转化被引量:2

RBX1蛋白是基于Cullin蛋白的E3复合体中的一个亚基,对于基于Cullin蛋白的E3复合体结合E2及Cullin蛋白的活化具有重要作用.利用RT-PCR分离到水稻RBX1cDNA,序列比对分析显示,水稻RBX1与拟南芥、人、果蝇及酵母中该同源蛋白的氨基酸序列一致性分别为80%、74·4%、72%和51·2%.酵母双杂交实验显示,水稻RBX1与水稻Cullin4蛋白有着相互作用.将水稻RBX1cDNA插入植物表达质粒pHB,利用农杆菌介导法将构建好的植物表达载体导入水稻,获得植株38株.以基因组DNA为模板对潮霉素抗性基因片段进行扩增,初步确定其中32株为转基因植株.

Alterations of Alternative Splicing Patterns of Ser/Arg-Rich (SR) Genes in Response to Hormones and Stresses Treatments in Different Ecotypes of Rice (Oryza sativa)被引量:5

Ser/Arg-rich (SR) genes encode proteins that play pivotal roles in both constitutive and alternative splicing of pre-mRNA. However, not much effort has been made to investigate the alternative splicing of their own pre-mRNA. In this study, we conducted comprehensive analyses of pre-mRNA splicing for 22 SR genes in three rice (Oryza sativa L.) ecotypes indica, japonica andjavanica. Using different ecotypes we characterized the variations in expression and splicing patterns of rice SR genes in different tissues and at different developmental stages. In addition, we compared the divergence in expression and splicing patterns of SR genes from seedlings of different rice ecotypes in response to hormones application and environmental stresses. Our results revealed the complexity of alternative splicing of SR genes in rice. The splicing varies in different tissues, in different ecotypes, in response to stresses and hormones. Thus, our study suggested that SR genes were subjected to sophisticated alternative splicing although their encoding proteins were involved in the splicing process.

A conserved unusual posttranscriptional processing mediated by short,direct repeated (SDR) sequences in plants

In several stress responsive gene loci of monocot cereal crops,we have previously identified an unusual posttranscriptional processing mediated by paired presence of short direct repeated (SDR) sequences at 5' and 3' splicing junctions that are distinct from conventional (U2/U12-type) splicing boundaries.By using the known SDR-containing sequences as probes,24 plant candidate genes involved in diverse functional pathways from both monocots and dicots that potentially possess SDR-mediated posttranscriptional processing were predicted in the GenBank database.The SDRs-mediated posttranscriptional processing events including cis-and trans-actions were experimentally detected in majority of the predicted candidates.Extensive sequence analysis demonstrates several types of SDR-associated splicing peculiarities including partial exon deletion,exon fragment repetition,exon fragment scrambling and trans-splicing that result in either loss of partial exon or unusual exonic sequence rearrangements within or between RNA molecules.In addition,we show that the paired presence of SDR is necessary but not sufficient in SDR-mediated splicing in transient expression and stable transformation systems.We also show prokaryote is incapable of SDR-mediated premRNA splicing.

玉米两个功能未知转录因子基因的转录后加工

ZmBH、ZmWD是玉米中分别具有转录因子bHLH和WD重复蛋白结构的两个功能未知基因,在不同组织中及各种胁迫诱导条件下,这两个基因均发生不同程度的剪接,产生大小各异的转录本.序列分析发现其剪接方式与经典的mRNA剪接不同,即剪接位点的选择不符合GT-AG或AT-AC规则,而是在剪接位点具有短正向重复序列SDR(Short-direct repeats).ZmBH的转录后加工导致序列发生移码而提前终止,而ZmWD则缺失了大部分结构域序列,经由这种方式产生的转录后加工产物可影响蛋白编码,因而可能是对这两个基因功能进行调控的一种方式.
