Diversity of microbial community at acid mine drainages from Dachang metals-rich mine, China被引量:3

Two acid mine drainage(AMD)samples TS and WK,which were from the Dachang metals-rich mine in Guangxi province,China,were studied using PCR-based cloning approach.A total of 44 operational taxonomic units(OTUs)were obtained from the two AMD samples.However,only three OTUs(GXDC-9,GXDC-19 and GXDC-50)detected in sample TS can also be observed in sample WK.Phylogenetic analysis revealed that the bacteria in the two samples fell into four putative divisions,which were Nitrospira,Alphaproteobacteria,Gamaproteobacteria,and Acidobacteria.Organisms of genuses Acidithiobacillus and Leptospirillum,which were in gamaproteobacteria class and Nitrospira family,were dominant in two samples,respectively.In sample TS,which was characterized by low pH,high sulfate,high iron,and high arsenide,two species(Acidithiobacillus ferrooxidans and Leptospirillum ferrooxidans)constituted 98.22% of the entire microbial community.Compared with sample TS,the microbial community in sample WK was more diversified according to the observation.Interestedly,the Legionella species,which was rarely observed in the low-pH environment,was detected in sample WK.This work helps us to further understand the diversity of microbial community living in extreme acid mine drainages with unique geochemistry and the tolerance capability of acidophiles to heavy metal.

氧化亚铁硫杆菌中铜代谢相关两个基因的差异(英文)被引量:2

目的:氧化亚铁硫杆菌(Acidithiobacillus ferrooxidans)在微生物冶金中发挥着重要的作用,研究其铜代谢机理有着十分典型的意义。在A.ferrooxidans全基因组序列数据库中,4个基因被注释与铜代谢相关。其中两个基因,Afe0454和Afe1073目前为止未发现有实验报道。本文旨在研究Afe0454和Afe1073与铜代谢的相关性。方法:通过一系列的方法如实时定量PCR、反转录PCR、序列分析,将基因导入抗铜基因缺陷的大肠杆菌(Escherichia coli)菌株中等,研究了Afe0454和Afe1073。结果:与Afe0454相比,Afe1073的表达对铜压力更敏感;Afe1073作为一个转录子单独转录,而Afe0454与Afe0453一起转录;序列分析显示Afe1073表达一种典型的重金属离子泵P1b1型ATP酶,而Afe0454表达一种未知功能的跨膜蛋白;不像Afe0454,Afe1073能使P1b型ATP酶敲除的大肠杆菌菌株铜抗性提高。结论:单独转录的Afe1073比Afe0454在铜代谢中发挥的作用更加明显。

Preservation of moderately thermophilic culture by freeze drying and frozen preservation way and effect on subsequent bioleaching of chalcopyrite被引量:2

Freeze drying and frozen preservation way was used to preserve a moderately thermophilic culture for bioleaching of chalcopyrite concentrate.After preservation of 15 months,the cell viability rate decreases to 22% with a cell density of 7×107 mL-1.When the growth time was extended from 8 days to 14 days,cell density would increase in a large scale to about 3×108 mL-1.In the bioleaching experiments,unpreserved and preserved cultures were compared for dissolving chalcopyrite concentrate.Before 44 days,the unpreserved culture can reach a high copper extraction of about 17.4 g/L.While the preserved culture shows a rather low copper extraction,which is only 9.7 g/L.When the bioleaching time was extended to 80 days,copper extraction by preserved culture increases remarkably,and the concentration of copper finally achieves up to 18.3 g/L.On the other hand,copper extraction by the unpreserved culture does not show remarkable increase from 44th to the 80th day,and finally the total copper extraction is 19.8 g/L.As a result,total copper extraction in 80 days by preserved culture approaches that by unpreserved culture and freeze drying and frozen preservation even after 15 months does not bring much decrease of bioleaching ability.

Repetitive sequence based polymerase chain reaction to differentiate close bacteria strains in acidic sites被引量:1

为了学习细菌的差异,紧张最新在中国从几酸矿排水( AMD )孤立地点,重复顺序基于的聚合酶链反应( rep-PCR ),为密切相关的细菌的差异分析的一种很好确定的技术拉紧,在细菌 Leptospirillum ferriphilium 的 30 紧张上被进行,细菌 Acidithiobacillus ferrooxidans 的 8 紧张,以及 Acidithiobacillus ferrooxidans 类型拉紧 ATCC (美国类型文化收集) 23270 。结果显示出那,用 ERIC 和盒子教材集合, rep-PCR 生产了高度差别对待的 banding 模式。基于 ERIC-PCR, banding 打的种系发生的分析被做,结果显示 rep-PCR 能在学习细菌的差异被用作一种快速、高度差别对待的屏蔽技术,特别在区分在在 AMD 的一种以内的细菌。
