红花黄色素抑制血小板激活因子诱导的内皮细胞炎性因子蛋白表达升高作用的研究被引量:11

目的:观察红花黄色素(SY)抑制血小板激活因子(PAF)诱发的内皮细胞炎性因子蛋白表达升高及抑制PAF与其受体特异性结合的作用。方法:用酶联免疫吸附测定法(ELISA)检测SY缓解1μmol/L PAF刺激脐静脉内皮细胞株Eahy926培养液中的炎症因子白细胞介素(IL)-1β、IL-6和肿瘤坏死因子(TNF)-α的蛋白表达水平的变化;以放射配基结合试验观察SY抑制[3H]标记的PAF与家兔血小板膜和膜蛋白PAF受体特异性结合的作用。结果:与正常组比较,PAF损伤组内皮细胞培养上清液中IL-1β、IL-6及TNF-α蛋白表达水平明显升高(P<0.001),SY处理组(浓度为4.5×10-5,4.5×10-4,4.5×10-3g/L)时可明显抑制Eahy926培养液中IL-1β、IL-6及TNF-α蛋白表达水平的升高(P<0.05,0.01,0.001);受体结合实验结果表明浓度为16.49g/L的SY可抑制0.17 nmol/L[3H]PAF与家兔血小板膜的特异性结合,浓度为11.17和22.08g/L的SY分别可抑制0.083及0.25nmol/L[3H]PAF与兔血小板膜蛋白的特异性结合。结论:SY通过抑制PAF与其受体的特异性结合缓解了PAF诱发的血管内皮细胞炎症介质IL-1β、IL-6及TNF-α蛋白的高表达。

红花是传统中药,具有活血化瘀功效。红花黄色素(SY)属查耳酮类化合物,是红花发挥治疗疾病作用的主要成分。羟基红花黄色素A(HSYA)为SY中含量最高的黄酮类成分。现代研究证实其对呼吸系统相关疾病都有明确的缓解和保护作用。SY及其主要单体成分HSYA有多种药理作用,本文对SY在呼吸系统肺部疾病中的一些药理作用进行综述。

羟基红花黄色素A抑制转化生长因子-β1诱导NIH/3T3细胞的活化作用被引量:2

目的:探讨羟基红花黄色素A(HSYA)抑制转化生长因子β1(TGF-β1)诱导NIH/3T3细胞活化的作用。方法:NIH/3T3细胞分8组:正常对照组、HSYA对照组、TGF-β1组、TGF-β1+0.5μmol/L HSYA组、TGF-β1+1μmol/L HSYA组、TGF-β1+2μmol/L HSYA组、TGF-1+2μmol/L SB-431542和TGF-β1+SB-431542+2μmol/L HSYA组。采用四甲基偶氮唑蓝(MTT)法检测细胞的增殖;细胞划痕实验观察细胞的迁移。结果:MTT法观察结果表明:与各时间点正常对照组相比,TGF-β1作用24,48及72小时后的OD 570nm值明显升高(均P<0.01),并且同时高于TGF-β1+HSYA各组(均P<0.05)和TGF-β1+SB-43154两组。在TGF-β1作用0,2,8,12及24小时后,与正常对照组相比,细胞迁移距离明显增加(均P<0.001),并且同时高于TGF-β1+HSYA组(均P<0.001)和SB-431542组。结论:HSYA可抑制TGF-β1诱导NIH/3T3细胞活化作用并且呈现时效关系和量效关系。

羟基红花黄色素A抑制转化生长因子-β1诱导的与肺纤维化相关信号通路的机制研究被引量:17

目的:探讨羟基红花黄色素A(HSYA)抑制转化生长因子-β1(transforming growth gactor-β1,TGF-β1)诱导的与肺纤维化相关信号通路的机制研究。方法:运用蛋白质印迹法(western blot)检测p38MAPK、Smad2磷酸化水平以及Smad2核转移情况。结果:HSYA能抑制TGF-β1组的p38MAPK、Smad2的磷酸化水平升高和Smad2的核转移,且高剂量HSYA组(16μmol/L)的抑制作用最明显(均P<0.05),此外,TGF-β1+si RNA和TGF-β1+16μmol/L HSYA+si RNA组的p38 MAPK、Smad2的磷酸化水平无明显差异,TGF-β1+SB-431542和TGF-β1+SB-431542+16μmol/L HSYA组的p38MAPK、Smad2的磷酸化水平以及Smad2的核转移情况均无明显差异。结论:HSYA可抑制TGF-β1诱导NIH/3T3细胞与肺纤维化相关的信号转导,呈现明显的量效关系,其作用靶点可能为TGF-βRII。

羟基红花黄色素A对香烟提取物诱导A549细胞炎症因子表达作用的研究被引量:6

目的:探讨羟基红花黄色素A(HSYA)抑制香烟烟雾提取物(CSE)诱导的A549肺泡上皮细胞炎症介质表达升高的作用。方法:用CSE刺激A549肺泡上皮细胞建立损伤模型,细胞分七组:单纯HSYA组、Sham组、CSE组、CSE+HSYA低剂量组(1μmol/L)、CSE+HSYA中剂量组(4μmol/L)、CSE+HSYA高剂量组(16μmol/L)和CSE+阳性对照药红霉素(5μg/mL)组。以RT-qPCR技术检测白介素(IL)-6、白介素(IL)-1β、肿瘤坏死因子(TNF)-α、细胞间黏附分子(ICAM)-1和血管细胞黏附分子(VCAM)-1 mRNA的表达水平;ELISA法检测细胞培养液中IL-6、IL-1β及TNF-α的蛋白水平表达;蛋白质印迹法(western blot)检测细胞p38MAPK磷酸化的水平。结果:与Sham组相比,CSE组IL-6、IL-1β、TNF-α、ICAM-1和VCAM-1 mRNA水平均明显升高(P<0.001),给予不同浓度的HSYA后炎症因子的高表达受到抑制,红霉素组也见较明显的抑制。与Sham组相比,CSE组细胞上清液中IL-6、IL-1β和TNF-α蛋白的表达水平明显升高(P<0.001),给予不同浓度的HSYA后CSE所诱发的炎症因子的高表达受到抑制,红霉素组也见较明显的抑制。与Sham组比较,CSE组的p38 MAPK的磷酸化水平明显升高(P<0.001),给予不同浓度的HSYA后细胞p38 MAPK的磷酸化水平升高受到抑制,红霉素组也可见明显的抑制。结论:HSYA可抑制CSE诱导A549细胞的炎症因子的表达升高。

Protective Effect of Hydroxysafflor Yellow A on Inflammatory Injury in Chronic Obstructive Pulmonary Disease Rats被引量:11

Objective: To investigate the attenuating effect of Hydroxysafflor yellow A(HSYA) on inflammatory injury in chronic obstructive pulmonary disease(COPD). Methods: Rats were randomly assigned to 7 groups according to body weight including normal control group, HSYA blank group(76.8 mg/kg), COPD group, COPD+HSYA(30, 48, 76.8 mg/kg) groups and COPD+dexamethasone(2 mg/kg), 10 in each group. Passive cigarette smoke and intratracheal instil ation of lipopolysaccharides were used to establish a COPD model in rats. Hematoxylin and eosin staining of lung tissue sections was used, real-time polymerase chain reaction(PCR) was used to assay m RNA levels of some cytokines in lung tissues, the cytokines in bronchoalveolar lavage fluid(BALF) were measured by enzyme-linked immunosorbent assay(ELISA), Western blot analysis was used to determine phosphorylated p38 mitogen-activated protein kinase(MAPK) levels in lung tissues, and nuclear factor-κB(NF-κB) p65 protein levels in lung tissues were detected by immunohistochemistry. Results: Lung alveolar septa destruction, alveolus fusion, inflammatory cel infiltration, and bronchiole exudation were observed. These pathological changes were al eviated in the COPD+HSYA group. The m RNA expression of inflammatory factors were significantly increased in lung tissues from COPD rats(all P<0.01) and were inhibited by HSYA. Levels of inflammatory cytokines in BALF of COPD rats were significantly increased(all P<0.01) which were inhibited by HSYA(all P<0.01, 48, 76.8 mg/kg). The levels of p38 MAPK phosphorylation and p65 in lung tissues of COPD rats were significantly increased(al P<0.01) and were suppressed by HSYA(all P<0.01, 48, 76.8 mg/kg). Conclusions: HSYA could alleviate inflammatory cell infiltration and other pathological changes in the lungs of COPD rats. HSYA could inhibit inflammatory cytokine expression, and increase phosphorylation of p38 MAPK and NF-κB p65 in the lungs of COPD rats. 羟基红花黄色素A药理作用及机制研究进展被引量:8

红花为菊科红花属植物红花的干燥花,是传统的活血化瘀类中药。本草纲目记载有“活血、润燥、止痛、散肿、通经”功效。临床上常用于闭经、痛经、产后恶露不行、跌打损伤、疮疡肿痛等。羟基红花黄色素A(HSYA)化学式为C27H32O16分子量612.5,是红花中主要活性成分,为一查尔酮葡萄糖苷。HSYA具有抗氧化、扩张冠状动脉、保护心肌等多种药理学作用,主治冠状动脉粥样硬化性心脏病、心绞痛、脑缺血再灌注损伤等心脑血管疾病。近年来,随着研究者对HSYA研究的不断深入,人们对其认识逐步深入,现将其药理作用及作用机制进行综述。

Objective:This study observed attenuating effect of hydroxysafflor yellow A(HSYA),an effective ingredient of aqueous extract of Carthamus tinctorius L,on lipopolysaccharide(LPS)-induced endothelium inflammatory injury.Methods:Eahy926 human endothelium cell(EC) line was used;thiazolyl blue tetrazolium bromide(MTT) test was assayed to observe the viability of EC;Luciferase reporter gene assay was applied to measure nuclear factor- κB(NF- κB) p65 subunit nuclear binding activity in EC;Western blot technology was used to monitor mitogen activated protein kinase(MAPKs) and NF- k B activation.Reverse transcription polymerase chain reaction(RT-PCR) method was applied to observe intercellular cell adhesion molecule-1(ICAM-1) and E-selectin mRNA level;EC surface ICAM-1 expression was measured with flow cytometry and leukocyte adhesion to EC was assayed with Rose Bengal spectrophotometry technology.Results:HSYA protected EC viability against LPS-induced injury(P<0.05).LPS-induced NF-κB p65 subunit DNA binding(P<0.01) and nuclear factor of kappa light polypeptide gene enhancer in B-cells inhibitor α(I κ Bα) phosphorylation was inhibited by HSYA.HSYA attenuated LPS triggered ICAM-1 and E-selectin mRNA levels elevation and phosphorylation of p38 MAPK or c-Jun N-terminal kinase MAPK.HSYA also inhibited LPS-induced cell surface ICAM-1 protein expression(P<0.01) and leukocyte adhesion to EC(P<0.05).Conclusion:HSYA is effective to protect LPS-induced high expression of endothelium adhesive molecule and inflammatory signal transduction.
