异基因造血干细胞移植后T辅助细胞亚群重建研究被引量:2

目的研究异基因造血干细胞移植(allo.HSCT)后CD4+T细胞及CD8+T细胞中CD25+Foxp3+(CD4+Treg细胞或CD8+Treg细胞)、IL一17a(Thl7细胞或Tcl7细胞)及IFN.7(Thl细胞或Tcl细胞)表达以及CD4+CD25。CDl27+效应性T细胞(Tcon细胞)重建,比较HLA相合与不合allo—HSCT后T细胞亚群重建的异同。方法收集2011年12月至2012年10月在北京大学血液病研究所进行allo—HSCT后未发生急性移植物抗宿主病的成年血液病患者20例,其中HLA相合10例、HLA不合10例,并留取10份健康供者外周血标本作为健康对照。采用8色流式细胞学检测技术检测CD4+T细胞及CD8+T细胞上CD25+Foxp3+、CD127、IL-17a及IFN-γ表达。结果@HLA全合组与不合组患者移植后2个月内CD3+T细胞、CD4+T细胞、CD8+T细胞数量重建两组问差异无统计学意义。②HLA相合组CD4+Treg细胞[+30d,8.46(0.36~27.41)细胞数/μl对1.10(0.04~8.03)细胞数/μl,P〈0.05;+60d,8.50(1.16-36.20)细胞数/μl对2.73(0.34~6.84)细胞数/μl,P〈0.05]、CD4+CD25-CD127+效应性T细胞[+30d,72.69(3.85~211.73)细胞数μl对13.41(0.48~96.17)细胞数/μl,P〈0.05;+60d,100.85(16.28~267.20)细胞数μl对47.75(6.34~143.04)细胞数/gl,P〈0.05]、Thl7细胞[+30d,2.34(0.02~6.87)细胞数/μl对0.20(0.02~1.34)细胞数/μl,P〈0.05;+60d,1.90(0.36~7.82)细胞数/μl对0.46(0.03~1.39)细胞数似l,P〈0.05]、Tcl7细胞数量(+30d,1.08(0.07~15.03)细胞数/μl对0.25(0.01~0.81)细胞数/pl,P〈0.05;+60d,1.85(0.63~26.57)细胞数仙l对0.46(0.01~3.66)细胞数仙l,P〈0.05]在+30d及+60d均明显高于HLA不合组,Thl细胞及Tcl细胞数量重建两组间差异无统计学意义。⑧Thl与Thl7细胞比值及T

单倍体相合造血干细胞移植后重建的淋巴细胞数量对受者预后的预测

目的探讨急性淋巴细胞白血病(ALL)患者接受非体外去除T淋巴细胞的单倍体相合骨髓和外周血造血干细胞移植(HBMT)后30d重建的淋巴细胞绝对数量(ALC-30)对移植预后的预测。方法回顾性分析2003年1月至2007年9月因ALL接受HBMT的62例受者的临床资料,根据受者的ALC-30水平,将受者分为ALC-30≥300/μl组(高ALC-30组)和ALC-30〈300/μl组(低ALC-30组)。观察两组受者移植后ALL复发、移植相关死亡、总体存活率(OS)和无复发存活率(LFS)。结果单因素及多因素分析结果均显示,与低ALC-30组相比,高ALC-30组受者具有较低的移植相关死亡率[(10.5±5.7)%和(30.8±8.1)%,(P=0.046)]和ALL复发率[(14.7±6.8)%和(59.1±10.3)%,(P=0.002)]及较高的OS[(79.3±7.5)%和(45.1±8.7)%,(P=0.006)]和LFS[(79.3±7.5)%和(28.4±8.0)%,(P〈0.001)]。结论ALL患者接受HBMT后30d淋巴细胞快速重建可以预测良好的移植预后。

An o-nitrobenzyl-based photocleavable antitumor prodrug with a terminal carboxyl group was designed and synthesized.The photolysis properties of the prodrug were investigated by means of1H NMR,HPLC,UV,and MTT methods.The results showed that the toxicity of the anticancer drug was effectively shielded before release.However,the prodrug effectively regained the antitumor capability against cancer cells by release of 5-fluorouracil when it was exposed to ultraviolet irradiation.

Synthesis and properties of novel photochromic bisthienylethenes containing imidazole and imidazolium derivatives被引量:1

A series of bisthienylethenes containing imidazole and imidazolium derivatives have been prepared and the products have been characterized by means of NMR and MS.Their photochromic and fluorescent switch properties have been investigated by UV–vis absorption spectra and fluorescence spectra.The fluorescent emissions of these kinds of photochromic compounds can be simply modulated by varying the imidazole groups,which shows that these compounds may have potential application in the design of fluorescent photochromic materials.

Synthesis, two-photon absorption and AIE properties of multibranched thiophene-based triphenylamine derivatives with triazine core被引量:3

Two new multibranched thiophene-based triarylamine derivatives with 1,3,5-triazine core are synthesized and characterized. Their one-and two-photon absorption properties and aggregation-induced emission effect have been investigated. Both the STAPA-based compounds are AIE active. The two-photon absorption (2PA) cross sections measured by the open aperture Z-scan technique are determined to be 620 and 1610 GM for STAPA-a and STAPA-b in chloroform,respectively, which dramatically increase with the introduction of alkyl chains. The relationship between their structures and properties on one-and two-photon absorption and aggregation-induced emission is discussed, which allows us to examine the effect of introducing alkyl chains. In addition, solvent effects also show a significant influence on the 2PA cross section. The two compounds with excellent AIE and 2PA properties provide attractive alternatives for the biophotonic materials.

A near-infrared fluorescent probe for monitoring fluvastatin-stimulated endogenous H_2S production被引量:3

Most reported fluorescent probes have limitations in practical applications in living systems due to the strong autofluorescence background,construction of probes with near-infrared(NIR) fluorescence emission is an accessible approach for addressing this challenge.We here designed a NIR fluorescent probe for monitoring the endogenous production of H_2S in living cells.The designed probe showed significant NIR fluorescence turn-on response to H_2S with high selectivity,enabling the sensitive detection H_2S.Importantly,the probe could be applied in monitoring the endogenous production of H_2S in raw 264.7 macrophages.This study showed that fluvastatin can promote the activity of cystathionineγ-lyase(CSE) for generation H_2S.
