Selection of Reference Genes in Saccharopolyspora Spinosa for Real-Time PCR

Reverse transcription quantitative PCR (RT-qPCR) combined with the published genome information of Saccharopolyspora spinosa can allow sophisticated studies about S. spinosa, including Studying the regulation of spinosyn biosynthesis, finding new target genes for engineering, and discovering and exploiting other macrolide secondary metabolites. Studies have demonstrated that appropriate internal control is needed to normalize target genes at transcription levels. However, many studies have shown that no single reference gene is universal for all strains under all experimental conditions. Thus, eight candidate reference genes of three different S. spinosa strains in two different cultures were studied to find suitable reference gene(sl. The number of amplification cycles of these candidate genes was calculated by BestKeeper, NormFinder and geNorm. The results indicated that the most suitable reference genes for normalization during the fermentation of S. spinosa were 16S rRNA and rbL13.

混合油脂补料发酵提高多杀菌素的产量被引量:7

通过补加植物油(菜籽油、玉米油、豆油)、油酸甲酯、玉米浆,考察其对多杀菌素发酵产量的影响。结果显示,在96h补加3%的混合油脂(油酸甲酯∶菜籽油=2∶1),同时在96h和144h分别补加1%的玉米浆可使多杀菌素的产量提高2.11倍,发酵单位达到804.59 mg/L,为实现工业化生产奠定基础。

Fed-Batch Fermentation for Spinosad Production in an Improved Reactor被引量:2

As a kind of aerobic bacteria, Saccharopolyspora spinosa exhibits a high demand for oxygen. In the fermentation process, the methods of increasing ventilation and improving agitation speed are usually adopted to achieve higher values of dissolved oxygen. These methods decrease the efficiency of spinosad biosynthesis. In this study, an improved reactor was designed to solve these problems. The exhaust gas reflux device, impellers, and baffles were improved. Furthermore, we established the kinetic models for the cell growth, substrate consumption and spinosad generation in batch fermentation process. The simulation results were in good agreement with the experimental data. Spinosad production reached 583.86 mg/L after employing the suitable feeding strategy by fed-batch fermentation in the improved reactor, whereas it was only 157.01 mg/L before optimization. The method described can provide insight to strengthen spinosad production and can be extended to the culturing process of filamentous aerobic bacteria. © 2017 Tianjin University and Springer-Verlag Berlin Heidelberg

异源表达聚酮类化合物前体的研究进展被引量:2

聚酮类抗生素因其重要的生物活性在临床上广泛应用。该类化合物主要由微生物代谢生成。然而由于合成聚酮化合物的天然菌株存在各种限制,影响着聚酮化合物的高效生产与应用。通过聚酮化合物的异源表达可以有效解除这些限制,目前成为研究热点。聚酮化合物异源表达的一个关键限制性因素是胞内前体物质供应不足。概述了在典型异源表达宿主菌中提高聚酮化合物前体供应的策略:通过对前体合成途径的重新改进或者引入到异源表达宿主菌中重构代谢网络、过表达外源前体合成途径、抑制旁支代谢流、敲除内源性次级代谢产物代谢途径,提高前体供应。为异源表达聚酮化合物的研究及工业化生产提供借鉴。
