Effect of Acupuncture on CXCL8 Receptors in Rats Suffering from Embryo Implantation Failure被引量:1

To observe the effect of acupuncture on CXCL8 receptors(CXCR1 and CXCR2)in rat endometrium experiencing embryo implantation failure,72 pregnant rats were randomly divided into four groups:normal group(N),embryo implantation failure group(M),acupuncture treatment group(A),and progestin treatment group(W).Then the rats in each group were equally randomized into a day-6(D6)group,a day-8(D8)group,and a day-10(D10)group.The rats in group M,group A,and group W were treated with mifepristone-sesame oil solution on day 1,while the rats in group N were injected with the same amount of sesame oil.Meanwhile,"Housanli"and"Sanyinjiao"were selected for acupuncture.From day 1 to the time of death,the rats in group A were fastened up and then acupuncture was administered while the rats in group N and group M were only fixed,and the rats in group W were given progestin.The number of implanted embryos was calculated.The expression of CXCR1 and CXCR2 in rat endometrium was detected by immunohistochemistry,Western blotting and real-time PCR.Compared to group N,the average number of implanted embryos,the protein and mRNA expression of CXCR1(D6,D8 and D10),and the protein and mRNA expression of CXCR2(D8 and D10)in rat endometrium were significantly decreased in group M.Compared to group M,there was significant elevation in the average number of implanted embryos,the protein expression(D6,D8 and D10)and mRNA expression(D8)of CXCR1 in rat endometrium of group A,and the protein expression(D8 and D10)and mRNA expression(D8)of CXCR2 in rat endometrium of group W.These findings indicated that acupuncture can increase the number of implanted embryos in rats of embryo implantation failure,which may be relevant with up-regulation the expression of CXCR1and CXCR2 at maternal-fetal interface of rats with embryo implantation failure.

针刺对胚胎着床障碍大鼠子宫内膜CCR2的影响被引量:10

目的观察针刺对胚胎着床障碍大鼠子宫内膜趋化因子受体CCR2表达的影响。方法将72只孕鼠随机分为正常组(N)、胚胎着床障碍组(M)、针刺治疗组(A)和黄体酮治疗组(W),每组随机再分为第6天组(D6)、第8天组(D8)、第10天组(D10)。其中M组、A组和W组大鼠妊娠第1天给予米非司酮麻油溶液造模,N组仅给予等量麻油溶液。同时,A组固定并每天针刺三阴交、后三里,N组和M组仅固定,W组肌肉注射黄体酮,直至处死。用免疫组织化学法、Western blot和Real-time PCR等方法测定着床点子宫内膜CCR2的表达。结果与N组相比,M组大鼠子宫内膜CCR2蛋白和mRNA的表达均明显降低,差异有统计学意义(P<0.05);与M组相比,A组和W组大鼠子宫内膜CCR2蛋白和mRNA的表达则明显增高(P<0.05)。结论针刺能上调胚胎着床障碍大鼠母胎界面CCR2的表达,从而促进胚胎的着床和胎盘的形成。

目的:观察针刺对胚胎着床障碍大鼠CD4+CD25+Foxp3+Treg细胞的影响。方法:144只孕鼠随机分为对照组(N)、米非司酮组(M)、米非司酮+针刺组(A)和米非司酮+黄体酮组(W),每组随机再分为6 d组、8 d组和10 d组。其中M组、A组和W组妊娠1 d给予米非司酮-麻油溶液造模,N组则给予等量麻油溶液。同时,每天下午A组固定并行针刺三阴交和后三里,N组和M组仅固定,W组肌肉注射黄体酮,直到处死当天结束。统计孕鼠胚胎着床数,用流式细胞术检测外周血CD4+CD25+Foxp3+Treg细胞和子宫内膜的CD4+Foxp3+Treg细胞的比例,Western blotting和real-time PCR测定着床点的子宫内膜Foxp3的表达。结果:与N组相比,M组的着床胚胎数、外周血CD4+CD25+Foxp3+Treg细胞和子宫内膜CD4+Foxp3+Treg细胞的比例、着床点子宫内膜Foxp3蛋白和mRNA的表达均明显下降(P<0.05);与M组相比,A组和W组的着床胚胎数、外周血CD4+CD25+Foxp3+Treg细胞和子宫内膜CD4+Foxp3+Treg细胞的比例以及着床点子宫内膜Foxp3蛋白和mRNA的表达均有不同程度地升高。结论:针刺改善胚胎着床障碍大鼠的胚胎着床可能与CD4+CD25+Foxp3+Treg细胞的调节密切相关。

Effect of Bushen Yiqi Huoxue Recipe on Placental Vasculature in Pregnant Rats with Fetal Growth Restriction Induced by Passive Smoking被引量:5

Interactions of vascular endothelial growth factor (VEGF) with receptors VEGFR1/Flt1 and VEGFR2/Flk1, and those of angiopoietins (Ang-1, Ang-2) with receptor Tie2 play important roles in placental angiogenesis. This study investigated vascular morphology and expression of these angiogenic factors in rat placenta on the day 15, 18, 21 of gestation (D15, D18 and D21). The rats were randomly assigned into 3 groups: normal group, model group [fetal growth restriction (FGR) model], and Bushen Yiqi Huoxue (BYHR) recipe treatment group (BYHR group, the pregnant rats with FGR were treated with BYHR recipe). Morphological analysis indicated that during initial villous formation, fetal nucleated erythrocytes (FNEs) appeared in maternal blood sinus (MBS). Subsequently, FNEs were surrounded by endothelial cells to form fetal capillary (FC) and then by trophoblast cells to form villi. As pregnancy proceeded, FC density increased progressively with increasing endothelial identification staining (EIS) in normal and BYHR groups. Whereas, villous formation was suppressed, normal increase in FC density was impaired and EIS was weakened in model group. Quantitative PCR analysis showed that VEGF and Flk1 mRNA increased over gestation in all groups, indicating that VEGF might play a pivotal role in FC growth during late gestation. VEGF mRNA was increased on D15, while decreased on D21 in model group as compared with normal group and BYHR group. Immunohistochemically, Ang-2 protein was highly expressed in FNEs, gradually disappeared as villi matured, and decreased over gestation in all groups, indicating that Ang-2 might play a pivotal role in villous formation, which was further supported by decreased Ang-2 mRNA and protein expression in model group on D15. Ang-1 mRNA, Tie2 mRNA and Ang-1/Ang-2 ratio increased from D15 to D18 in all groups as placenta matured. 被动吸烟诱导的胎儿生长受限大鼠胎盘血管结构异常及其机制被引量:2

目的研究被动吸烟诱导的胎儿生长受限(fetal growth retardation,FGR)大鼠胎盘血管结构的异常及其分子机制。方法采用被动吸烟法建立大鼠FGR模型,将12只孕鼠随机分为正常组(n=6)和FGR组(n=6),每只孕鼠取3个胎盘检测(即每组n=18)。运用凝集素标记血管内皮细胞以观察胎盘血管形态,体视学方法分析胎盘血管密度,实时荧光PCR检测胎盘血管因子VEGF-A、VEGFR1、VEGFR2、Tie2、Angpt1、Angpt2mRNA的表达,免疫组化及Westernblot检测VEGF-A、VEGFR2、Tie2、Angpt2蛋白的表达。结果 FGR组胎盘胎儿血管总体积为(45±21)mm3,显著小于正常组(70±19)mm3(P<0.05)。FGR组胎盘血液交换屏障厚度为(5.839±0.205)μm显著高于正常组(1.967±0.543)μm(P<0.05)。FGR组胎盘血管体积密度为(0.100±0.450)(0.032~0.158),正常组胎盘血管体积密度为(0.113±0.018)(0.086~0.135),两组间差异无统计学意义。与正常组相比,FGR组的VEGF-A mRNA表达显著下降(P<0.05),其它因子的mRNA表达较正常组微升高,但差异无统计学意义。FGR组的VEGF-A、VEGFR2、Tie2、An-gpt2蛋白表达有增加趋势,但差异无统计学意义。结论被动吸烟可使胎盘血管总体积显著减少、胎盘血液交换屏障厚度显著增加,导致胎盘营养交换功能不足,从而引发胎儿生长受限。但部分被动吸烟所诱导的FGR胎盘血管密度发生代偿性增加,以致FGR胎盘血管密度及血管因子mRNA、蛋白表达的改变不显著。

健胎液对围着床期小鼠子宫炎性细胞因子IL-1的影响被引量:3

目的:探讨健胎液对胚胎着床障碍小鼠着床期子宫炎性细胞因子IL-1β及IL-1RⅠ的影响。方法:建立胚胎着床障碍的动物模型,分为三组,观察各组着床率和着床点数,妊娠第5天分别检测各组小鼠子宫内膜IL-1β的蛋白含量,以及子宫内膜IL-1β、IL-1RⅠ的mRNA水平。结果:模型组胚胎着床数、着床位点数与正常组比较显著降低(P<0.01);中药组胚胎着床数、着床位点数与模型组比较明显升高(P<0.05),但仍低于正常组(P<0.05)。模型组子宫内膜IL-1β蛋白量同正常组、中药组比较明显升高,中药组IL-1β蛋白量同正常组比较无显著性差异。模型组IL-1β、IL-1RⅠ的mRNA表达水平同正常组比较显著升高,中药组IL-1RⅠ的mRNA表达水平同模型组比较明显降低,但仍比正常组水平高。结论:围着床期子宫内膜IL-1β、IL-1RⅠ参与胚胎着床的病理生理过程,健胎液可能通过调节IL-1表达,改善子宫内膜微环境,提高子宫内膜胚胎种植容受性,促进着床。
