一种基于纳米金介导生物沉积铂催化氢还原的电化学免疫分析新方法被引量:4

发展了一种基于纳米金介导生物沉积铂并以铂催化氢还原伏安法进行检测的高灵敏电化学免疫分析新方法.该方法采用夹心免疫分析模式,实现了人免疫球蛋白(hIgG)的测定.首先在聚苯乙烯微孔板中固定羊抗人hIgG捕获抗体,hIgG捕获后,碱性磷酸酶标记的hIgG抗体修饰的纳米金探针通过与hIgG形成夹心复合物而结合在微孔板上.结合的碱性磷酸酶催化抗坏血酸磷酸酯底物水解产生抗坏血酸,后者在纳米金的介导下还原铂离子沉积于纳米金表面.沉积的金属铂用王水[V(HNO3)∶V(HCl)=1∶3]溶解并电富集于玻碳电极上.通过测定铂催化氢还原产生的阴极电流,可实现hIgG的高灵敏分析.催化氢还原电流与hIgG浓度对数在100ng/mL~2μg/mL及100pg/mL~100ng/mL之间呈线性相关性,检测限达22pg/mL.由于铂催化氢还原的高灵敏度及纳米金介导的生物沉积放大反应,该法具有较高的分析灵敏度,且免疫分析微孔板模式使得该法可同时用于大量样品的分析.

Ultra-large Scale Molecular Dynamics Simulation for Nano-engineering被引量:3

In light of the special need of nano-engineering, an ultra-large scale and high-performance molecular dynamics(MD) simulation program was implemented. In many nano-engineering processes, the free boundary condition should be adopted. To meet this particular requirement, a pointer link and dynamic array data structures were employed so that both reliability and accuracy of simulation could be ensured. Using this method, one could realize the MD simulation of the nano-engineering system consisting of several million atoms per single CPU.

Electrochemical Detection of Clenbuterol in Pig Liver at Pyrrole DNA Modified Boron-doped Diamond Electrode

The direct detection of clenbuterol(CL) in pig liver without any extraction separation at a pyrrole-DNA modified boron-doped diamond(BDD) electrode is reported. The pyrrole-DNA modified BDD electrode has a strong electrocatalytic effect on the redox reaction of CL. One oxidization and two reduction peaks of CL appear at 340.2, 299.8 and 166.6 mV(versus SCE), respectively. The pyrrole polymer alone cannot electrocatalyze the above reaction at a BDD electrode; the electrocatalytic effect of a BDD electrode modified with DNA membrane is unsufficient for the analytical detection of CL; the replacement of boron-doped diamond by glass carbon makes the electrocatalytic reaction impossible; the redox process is pH dependent. The influences of various experimental parameters on the pyrrole-DNA modified BDD electrode were investigated. A sensitive cyclic voltammetric response for CL was obtained in a linear range from 3.4×10^-6 to 5×10^-4 mol/L with a detection limit of 8.5×10^-7 mol/L. A mean recovery of 102.7% of CL in the pig liver sample solution and a reproducibility of 3.2% were obtained.
