高效液相色谱法测定小柴胡汤中黄芩苷、汉黄芩苷的含量被引量:13

目的采用高效液相色谱法测定小柴胡汤中黄芩苷、汉黄芩苷的含量。方法饮片粉末用水回流提取,色谱柱:Agilent Zorbax XDB-C18(150mm×4.6mm,5μm);流动相为乙腈和水;A相为0.2%磷酸-水,B相为乙腈,梯度洗脱;流速1ml/min,柱温25℃,检测波长275nm,进样量15μl。结果黄芩苷和汉黄芩苷在30min内基线分离。以峰面积(Y)对浓度(X)的标准曲线分别为:黄芩苷Y=44.16X-36.22(r=0.9999);汉黄芩苷Y=52.08X-28.69(r=0.9999)。日内及日间精密度均小于1%,黄芩苷和汉黄芩苷的定量限分别为0.6156μg/ml和0.2208μg/ml,黄芩苷和汉黄芩苷加样回收率(n=6)分别为95.73%(RSD=0.8%)及97.02%(RSD=1.56%)。结论该方法稳定性好,测定结果准确可靠,可用于小柴胡汤的质量控制研究。

Comparative analysis of essential oils found in Rhizomes Curcumae and Radix Curcumae by gas chromatography-mass spectrometry被引量:5

A comparison of the volatile compounds in Rhizomes Curcumae (Ezhu) and Radix Curcumae (Yujin) was undertaken using gas chromatography-mass spectrometry (GC-MS).Ultrasonic extraction and GC-MS methods were developed for the simultaneous determination of five sesquiterpenes,namely,a-pinene,b-elemene,curcumol,germacrone and curdione,in Ezhu and Yunjin.Good linearity (r40.999) and high inter-day precision were observed over the investigated concentration ranges.The validated method was successfully used for the simultaneous determination of five sesquiterpenes in Ezhu and Yujin.The quantitative method can be effectively used to evaluate and monitor the quality of Chinese curcuma in clinical use.

Rapid analysis of components in Rhizoma Anemarrhenae by HPLC-DAD-MS and HPLC-DAD-TOFMS

A global quality control method based on high performance liquid chromatography(HPLC)coupled with diode array detection(DAD),single quadrupole mass spectrometry(MS)and time-of-flight mass spectrometry(TOFMS)was developed for simultaneous determination of seven major components(mangiferin,neomangiferin,timosaponin E1,timosaponin E,timosaponin BⅡ,timosaponin BⅢ,and timosaponin AⅢ)and identification of most components in extracts of Rhizoma Anemarrhenae(RA).HPLC analysis was performed on an Agilent SB-C18 column(4.6 mm×150 mm,5 μm)by gradient elution using acetonitrile and water-acetic acid(100∶0.05,v/v)as the mobile phase.Seven major components in RA were successfully separated.This quantitative method was fully validated in respect of the following performance criteria:linearity,precision,repeatability,stability,accuracy,limits of detection(LOD)and quantification(LOQ).A formula database of known compounds in RA was established,against which,most of the reported components in this herbal extract were identified effectively based on the extract masses acquired by TOFMS.This qualitative and quantitative method was successfully used to analyze the components in 10 batches of RA samples collected from different regions in China.This global quality control method,which consisted of HPLC-DAD-MS assay of seven major components and unambiguous identification of nineteen components,is suitable for routine quantification and comprehensive quality control of RA.

目的综述微透析与液相色谱-质谱联用技术在药物体内化学成分分析中的研究进展。方法参阅近几年国内外相关文献,对微透析与液相色谱-质谱联用技术在药物体内内源性物质及外源性物质中的应用进行归纳与总结。结果与结论微透析与液相色谱-质谱联用作为一种新型的分析技术,具有快速简便、干扰少、前处理简单等优点。此项技术在体内药物分析中得到了迅速推广和应用,特别在药代动力学研究中显示出其独特的优势,具有十分广阔的应用前景。

目的:研究莪术中的挥发性化学成分。方法:采用Headspace-GC/MS方法,TR-5石英毛细管色谱柱(30m×0.25 mm×0.25μm),顶空瓶平衡温度90℃,平衡时间30 min,进样量1.5 mL。结果:运用GC-MS能快速鉴定出79种化学成分。结论:本方法灵敏、快速、准确,为莪术挥发性成分研究提供了理论依据。

中药复方四逆汤化学成分的RRLC-TOF/MS分析被引量:11

目的:通过高分离度快速液相-飞行时间质谱(RRLC-TOF/MS)联用技术定性分析中药复方四逆汤中的主要化学成分。方法:色谱分离采用资生堂CAPCELL PAK C18反相柱(50 mm×2.0 mm,2μm),流动相组成分别为0.1%甲酸水溶液和乙腈,梯度洗脱,流速为0.25 mL/m in;质谱定性采用飞行时间质谱,正离子模式扫描。结果:在优化的液质联用条件下,通过飞行时间质谱鉴定出四逆汤中34个成分,并对其药材来源进行了归属。结论:通过RRLC-TOF/MS联用技术,为鉴定四逆汤中的化学成分建立起了一种快速、高效的分析方法。

目的:用高效液相色谱-飞行时间质谱(HPLC-TOF-MS)联用技术对荷丹片中化学成分进行鉴别与结构解析。方法:色谱分离采用Agilent Zorbax SB-Aq柱(250mm×4.6mm,5μm),流动相为乙腈(A相)-1%乙酸水溶液(B相),梯度洗脱:0~50min,0%~30%B相,50~90min,30%~75%B相;流速为1ml/min。质谱定性分析采用TOF-MS,在正离子模式下进行扫描,参比离子m/z分别为121.05和922.01。结果:在液质联用条件下,通过TOF-MS以及碎片离子裂解分析,鉴定了荷丹片中的33种化学成分和5组同分异构体。结论:HPLC-TOF-MS联用技术是鉴定荷丹片中化学成分的一种简单、有效的方法。

Using cell membrane chromatography and HPLC-TOF/MS method for in vivo study of active components from roots of Aconitum carmichaeli被引量:4

An offline two-dimensional system combining a rat cardiac muscle cell membrane chromatography time-of-flight mass spectrometry(CMC-TOF/MS)with a high performance liquid chromatography time-of-flight mass spectrometry(HPLC-TOF/MS)was established for investigating the parent components and metabolites in rat urine samples after administration of the roots of Aconitum carmichaeli.On the basis of the analysis of the first dimension,retention components of the urine sample were collected into 30 fractions(one fraction per minute).Then offline analysis of the second dimension was carried out.34 compounds including 24 parent alkaloids and 10 potential metabolites were identified from the dosed rat urine,and then binding affinities of different compounds on cell membranes were compared and influences of some functional groups on activity were estimated with the semi-quantification and curve fitting method.As a result,binding affinities decreased along with the process of deacylation,debenzoylation and demethylation,which may be related to the alleviation of toxicity in the procedure of herb processing or metabolism.Moreover,some minor components in rat urine(Songorine,14-benzoylneoline,Deoxyaconitine,etc.)exerted relatively strong affinity on cell membranes are worth exploring.The results delivered by the system suggest that the CMC can be applied to in vivo study.
