Catalyzed Resolutionb racemic 2-methyl-1-butanol was achieved,with max.95%optical purity of chiral alcohol,by candida cycindrace lipase in non-aqueous organic media.Effect of some factors on the resolution was investgated.The factors included temperature,pH,organic solvents,the amount of added water and the reaction time.
DL-2-methyl-1-butanol was resoluted by porcine pancreatic lipase(PPL)with tributyrin served as substrate and organic phase.Tributyrin,2-methylbutyl butyrate and dibutyrin in the sample from the reaction process were separated by TLC.Their respective spots were obtained by using developing solution of hexane:ether:ethyl-acetate:methanol(10:2:1:1 in vol.)and placing spreaded gel panel in iodine steam.The spots were analyzed by a TLC analyzing software designed by Visual C++programming language,with satisfactory accuracy.It shows that monitoring the reaction by TLC is realizable.
Kinetic resolution of(R,S)-2-methyl-1-butanol was completed by porcine pancreatic lipase-catalyzed transesterification with triacetin in a solvent-free system.The effect of reaction conditions such as solvents,water,pH,temperature,and the molar ratio of the substrates on lipase activity and stereoselectivity were examined.The results demonstrate triacetin has an advantage of low price and high activity when it is employed as the substrate in the kinetic resolution of(R,S)-2-mentyl-1-butanol and is a very promising tributyrin substitute for industrial application.
