长江江豚和瓶鼻海豚的血清转铁蛋白

Serum transferrins(Tf)of bottlenose dolphin(Tursiops truncatus)and Yangtze finless porpoise(Neophocaena phocaenoides asiaeorientalis)were analyzed by polyacrylamide gel electrophoresis(PAGE),and the molecular weight of bottlenose dolphin Tf was determined by sodium dodecylsulfate polyacrylamide gel electrophoresis(SDS-PAGE).Bottlenose dolphin Tf and Yangtze finless porpoise Tf appeared to be β-globulins.Both species have five bands even though they belong to different families,which is more bands than previously reported for some fish and terrestrial mammals.The distance between the Tf bands was gradually less from the fastest band to the slowest band.The last two bands were close to each other.However,there were no difference among the two bottlenose dolphins and three finless porpoises.Bottlenose dolphin Tf is composed of two components,whose molecular weight disparity is about 3 000 Da.The Tf molecular weights from two bottlenose dolphins were 72 700 Da/69 900 Da and 73 100 Da/69 700 Da,respectively.These values are obviously lower than those in previous studies of humans,rabbits,rats,Guinea pigs,horses,dogs,mice and cats.

厦门海域两头中华白海豚体内铁的含量

Samples of liver,pancreas,stomach,kidney,intestine,lung,muscle,heart,skin,and brain from two stranded Indo-Pacific humpback dolphins(Sousa chinensis)near Xiamen harbor of China in April 2004 were analyzed for iron(Fe)concentration.The highest levels of Fe were found in the blood(2 471.0 μg/g,dry weight)and liver(2 968.2 μg/g,dry weight)of the animal XM20040429(female,body weight 86 kg,body length1.88 m,estimated age 2-3 year-old)and XM20040430(female,204 kg,2.45 m,4-5 year-old),respectively.In general,Fe concentrations were higher in blood,liver,lung,kidney and muscle than any other organs or tissues in both individuals,although there were obvious differences in body length,body weight and age between these two dolphins.This tissular distribution pattern of Fe concentration of Indo-Pacific humpback dolphins in the present study is similar to those of Dall’s porpoises(Phocoenoides dalli),finless porpoises(Neophocaena phocaenoides)and striped dolphins(Stenella coeruleoalba)in the previous literatures.

Serum transferrin(Tf) is an iron(Fe)-transporting protein with the property of reversibility in binding Fe.Cetaceans are aquatic mammals that exhibit particularly high Fe bioavailability in the body.However,little is known about serum Tf in cetaceans to date.The goal of this study was to determine the special characters of serum Tf between cetacean species and several other vertebrate groups.Zones and types of serum Tf of cetacean bottlenose dolphin(Tursiops truncatus) and rough-toothed dolphin(Steno bredanensis) were studied comparatively,for the first time,with bottom-living yellow catfish(Pelteobagrus fulvidraco),sharptooth catfish(Clarias leather),turbot(Scophthalmus maximus),margined bullhead(Liobagrus marginatus),Chinese sturgeon(Acipenser sinensis),amphibian tiger frog(Rana tigrina),terrestrial mice(Mus musculus) and human(Homo sapiens).Electrophoretic separation of Tf was performed using discontinuous polyacrylamide gel electrophoresis(PAGE).An iron-specific stain method was used to identify the Tf and other Fe-binding proteins.Results suggested that the Tf zones were divided into four areas(Tf I-Tf IV).Serum Tf possessed 4-6 bands in the bottlenose dolphins and 5 Tf bands in the rough-toothed dolphin.These two Tf types in cetacean species were usually more complicated than those of human and other animals studied except for tiger frog.In addition,cetacean serum Tf distributed more widely across the serum Tf zones,and especially all serum Tf I-Tf IV zones in the bottlenose dolphin.The concentrative areas of serum Tf bands and their relative activities seemed to be different between cetaceans and other species,implying an adaptation for special water living habitats and respiratory patterns.Furthermore,although the serum Tf zone patterns among the three bottlenose dolphins were the same,the Tf bands were different from each other,suggesting the tendency of serum polymorphisms in serum Tf of the three bottlenose dolphins.Further research will be required to deepen our understanding not only of the cetacean

厦门海域两头中华白海豚体内微量元素的积累被引量:2

对2004年4月在厦门海域搁浅死亡的两头中华白海豚(Sousa chinensis)进行了肝脏、胰、胃、肾、肠、肺、肌肉、心脏等多组织微量元素锌(Zn)、铜(Cu)、汞(Hg)、铅(Pb)、镉(Cd)和砷(As)浓度的测定。两豚各组织总体上必需元素Zn、Cu浓度高于有毒元素Hg、Pb、Cd和As。XM20040430体内特别高的Zn、Hg、Pb、Cd浓度分别出现于皮肤(500μg/g干重)、肝脏(255μg/g干重)、肺(13.0μg/g干重)和肾(2.82μg/g干重)中,而Cu和As在各组织中的浓度较为相似。XM20040429体内仅肝脏中Zn(448μg/g干重)和Cu(52.0μg/g干重)的浓度远高于其他组织。虽然两海豚微量元素的总体积累多低于急性毒性水平,但有毒元素Hg、Pb、Cd、As的明显检出和XM20040429肝脏中异常高的Zn浓度均显示出进一步研究微量元素慢性胁迫效应的必要性。结果还显示厦门港海域近年来Hg、Cd、Pb污染未见减轻,As也明显检出,必须引起高度重视。今后有关水生哺乳动物中As积累的研究也有待加强。

鲇、大口鲇及其杂交鲇(鲇♀×大口鲇♂)血液生理生化指标比较及转铁蛋白等位基因分析被引量:3

分别测定了鲇(Silurus asotus Linnaeus)、大口鲇(S.meridionalis Chen)以及杂交鲇(鲇♀×大口鲇♂)的红细胞、白细胞、血小板、红细胞平均血红蛋白浓度、每升红细胞平均血红蛋白浓度以及血红蛋白等6个血细胞指标和血清铁、总铁结合力、未饱和铁结合力以及铁饱和度等4个血清铁指标,结果表明杂交鲇在呼吸功能上具有较强的优势;鲇、大口鲇以及杂交鲇血清转铁蛋白的PAGE分析表明:杂交鲇Tf表现型种类多、遗传均一性较好;杂交鲇血清Tf等位基因a、b和c分别来自大口鲇血清Tf等位基因a、鲇血清Tf等位基因a以及鲇和大口鲇的血清Tf共有的等位基因b。由此推断,杂交鲇比其亲本的耐低氧能力更强。

从长江上游水系采集到野生白缘(鱼央)。血清转铁蛋白 PAGE 分析显示:白缘(鱼央)血清转铁蛋白分两个区,即 Tfa 区和 Tfb 区,其中 Tfa 区有一条带,Tfb 区有两条带;本试验发现5个表现型,频率分别为34.78%,47.83%,4.35%,4.35%和8.69%,等位基因 a、b_1和 b_2的基因频率分别为37.74%、41.51%和20.75%;对白缘(鱼央)、黄颡鱼、革胡子鲇、中华鲟、大菱鲆、黑斑蛙、金环蛇、鹅、人、小鼠、牛、宽吻海豚以及糙齿海豚等13个物种的血清转铁蛋白分析表明:脊椎动物血清转铁蛋白 PAGE 图谱分为 Tfa、Tfb、Tfc 和 Tfd 4个区。
